B11 Metabolic Network Dynamics for Production of Recombinant Glycosyltransferases

The production of glycosyltransferases in Aspergillus niger and Bacillus megaterium will be investigated and optimized on the metabolic network level. Here, we focus on the carbon core metabolism due its key role in the synthesis of the target proteins. The planned studies will utilize novel concepts of 13C-based metabolic flux analysis for the quantification of small molecule fluxes, i.e. in vivo enzyme and pathway activities.

In close interaction with projects in areas A and B this will provide detailed insights into metabolic function and regulation towards identification of bottlenecks as a basis for rational improvement of production strains and processes. In addition the project will deliver key data for systems oriented modelling. Hereby, the fluxome displays an important link to the on-going SFB research on proteome, transcriptome and physiological phenotype.

The planned studies with B. megaterium will focus on novel concepts to analyze dynamic, production relevant processes, such as high cell density cultivations. For the eukaryote A. niger, only poorly studied on the flux level so far, the method will be established for defined conditions including the development of a suitable production medium and applied to compare the influence of environmental or genetic perturbation. Of specific interest here is the interrelation of morphology, metabolism and production behaviour.
