A6 Production of recombinant Antibodies in Bacillus megaterium and Aspergillus niger

Recombinant Antibodies are the active ingredients of the largest and also the fastest growing group of protein therapeutics. This growth results in an enormous demand for recombinant protein mass production. The worldwide manufacturing capacity is limited and it can be doubted that the health systems can support the use this dramatically increased number of expensive recombinant protein drugs in the future. In this project, alternatives to the common cost-intensive production in mamalian cells will be developed.

The main objective of this subproject ist to evaluate production hosts which promise to combine the advantages of microbes with the performance of mammalian cells. Well established systems, as E. coli, cannot be used, as this bacterium is not able to produce complete immunoglobulins. The filamentous fungus Aspergillus niger and the gram positive bacterium Bacillus megaterium are known to be able to secrete large and complex proteins into the growth medium. By improving vectors, strains and conditions for the production of various recombinant antibodies and their fusion proteins, recombinant strains for production and purification in Batch- or continuous operation will be created.
